Library construction
IIT Biotech prepare custom libraries of genomic DNA or BAC DNA from different bacteria or viruses. The template DNA has to be purified according to the IIT Biotech guidelines. Please notice that the material has to be classified as harmless. In case of BAC or Fosmid templates extraction by IIT Biotech is available. Shotgun libraries
Shotgun libraries are constructed from BAC or Fosmid DNA. Inserts will be cloned into standard cloning vectors. The strategy will be discussed with the customer.
Fosmid libraries
Fosmid libraries are constructed from pure whole genomic DNA. The inserts will be cloned into the fosmid vector pEPIFOS. These commonly used vectors are capable of carrying large regions of genomic DNA in E. coli. Fosmids are similar to cosmid vectors in that they contain cos sites for lamda packaging of cloned DNA inserts (35-45 kb). Like BACs, fosmids are derived from genetic elements of the single copy F factor, which enables fosmids to maintain genomic DNA inserts with higher stability than cosmids. Direct sequencing is one of the main downstream applications for the DNA of fosmids. The DNA can be used to obtain the genomic sequences at the ends of the cloned insert by using primers that anneal to the DNA at the vector-insert junction and reading into the cloned insert. These end sequences provide researchers with information that is used to map the clones and assemble the data for genome sequencing projects.
